چکیده

The etiology of neuronal death in neurodegenerative diseases, including Huntington′s disease (HD), is still unknown. There could be a complex interplay between altered energy metabolism, excitotoxicity and oxidative stress. Our aim was to examine the effects of intrastriatal injection of a selective inhibitor of neuronal nitric oxide synthase, 7 nitro indazole (7-NI), and non-specific potent nitric oxide synthase inhibitor, Nω-nitro-l-arginine methyl ester (lNAME), to investigate the possible involvement of nitric oxide in quinolinic acid (QA)-induced striatal toxicity in the rat. Unilateral administration of QA in rat striatum in a single dose of 150 nM was used as model of HD. The second and third group were treated with 7-NI and QA and l-NAME and QA, respectively. The control group was treated with 0.9% saline solution likewise. The content of reduced glutathione, a key antioxidant, was increased in the hippocampus and basal forebrain of 7-NIand QA-treated animals, compared to the QA-treated animals. These results support the hypothesis that oxygen free radicals contribute to the excitotoxic neuronal injury, and also that neuronal nitric oxide synthase inhibitor 7-NI, but not l-NAME could be potential neuroprotective agents in HD.

Improvement of Tissue Survival of Skin Flaps by 5α-Reductase Inhibitors: Possible Involvement of Nitric Oxide and Inducible Nitric Oxide Synthase

Background: Skin flap grafting is a popular approach for reconstruction of critical skin and underlying soft tissue injuries. In a previous study, we demonstrated the beneficial effects of two 5α-reductase inhibitors, azelaic acid and finasteride, on tissue survival in a rat model of skin flap grafting. In the current study, we investigated the involvement of nitric oxide and inducible nitric o...

Ameliorating of Memory Impairment and Apoptosis in Amyloid β-Injected Rats Via Inhibition of Nitric Oxide Synthase: Possible Participation of Autophagy

It has been proposed that appearance of amyloid beta (Aβ) in hippocampus is one of the characteristic features of Alzheimer’s disease (AD). The role of Nitric oxide (NO) in neurodegenerative disorders is controversy in different contexts. Here, we examined the effect of NO on spatial memory. For this purpose, we compared the effects of three different concentrations of L-NG-Nitroarginine Methyl...

Nitric Oxide is Protective Against Mercury Induced Depression

A B S T R A C T Introduction: Mercury is the second most metal pollutant in the world and has the potential to induce many pathologic conditions, especially in nervous system, such as depression. Here we tried to find out if nitric oxide has any possible role in the pathophysiology of depression induced by this metal. Although the role of nitric oxide has been shown in mood control, here we use...

The Involvement of Nitric Oxide in Antidepressant-Like Effect of Metformin after Bile-Duct Ligation in NMRI Mice

Background: In some disorders such as diabetes mellitus patients can display depressive symptoms. Metformin is among the first-line treatments for management of the type 2 diabetes mellitus which may have some anti-depressant effect. Objective: Current investigation was performed to examine the anti-depressant effects of metformin and the involvement of nitric oxide (NO) in this way, in an expe...
